The 27-year-old has signed up for a third spell at Old Trafford. In 2011 he took 12 wickets in eight T20 matches while in 2014 he took 19 in 10 at an average of 14, helping the Lancashire reach Finals Day in both campaigns.
Khan, who played for Middlesex in the Royal London Cup in 2015, will be available throughout the competition for Lancashire.
"I thoroughly enjoyed both my spells at Emirates Old Trafford and I'm looking forward to linking up with the squad later this summer," Khan said.
