Sunday's action kicks off at the DW Stadium as Sky Bet League One promotion hopefuls Wigan Athletic host Premier League strugglers Southampton.
The Latics will come into this one full of confidence after knocking out Manchester City in the last round, just the third team to beat Pep Guardiola's men in any competition this season.
Paul Cook's side also, as expected, find themselves in the automatic promotion spots in the third tier after a last-minute winner away to Bradford City on Wednesday night.
Mauricio Pellegrino's reign as Southampton boss was brought to an end this week after he was dismissed following defeat to Newcastle. Former Stoke boss, and Saints player, Mark Hughes comes in with the task of keeping the south coast side in the Premier League.
For Southampton, they'll be looking for a new manager bounce as they aim to improve a run that has seen just one win in five games.
These two sides last met in 2013 when Wigan were in the Premier League. Despite relegation that season, Athletic would go onto lift the FA Cup at Wembley, against beating Manchester City by a single goal.
With the difference in leagues, the hosts come into this one as the 13/5 outsiders, whilst Southampton can be found as short as 23/20 to secure passage into the next round.
The big question will be how much of a distraction will the FA Cup be for these two sides given their current respective league positions?
The Saints are 15/8 to be relegated from the top flight this season and that will be their priority despite being in the latter stages of the Cup.

Leicester v Chelsea (1630 GMT, BBC One)
Sunday's second football offering is an all Premier League affair as Leicester City host Chelsea.
Antonio Conte's Chelsea side were knocked out of the Champions League in midweek with a Lionel Messi masterclass on show in a 3-0 victory for Barcelona.
The Foxes will be boosted with extra rest time, having not played since last Saturday's 4-1 hammering of all-but-relegated West Brom at the Hawthorns.
Claude Puel's side will also be looking to extend their unbeaten run at home. A penalty shootout defeat to Manchester City in the Carabao Cup being the last time that they were defeated at the King Power Stadium.
For Chelsea, there is extra incentive as this is the only major trophy left for them to win. The Premier League, Champions League and Carabao Cup won't be in their trophy cabinet at the end of the season.
Chelsea can be found as 6/5 favourites despite being away, with Leicester at 13/5 and the draw around the 5/2 mark.
Much like Wigan and Southampton, this has the potential to go past 90 minutes as both sides aim to make the Wembley semi-final. Leicester's home record, combined with Chelsea's dismal away form may tilt the tie in the hosts' favour.
Conte will be eager to finish his time in England with a trophy, as there are big questions surrounding his future at the club, and they find themselves potentially just three wins away from doing so.
When it comes to looking for value, corners may be the market to explore. These two sides sit within the top nine of the Premier League sides with a high amount of corners in their games, Leicester find themselves in third.
