Frenchwoman Cornet has been having a good season, but had lost all three of her previous meetings with world number one Serena.
She held her nerve impressively this time, though, serving out her 6-4 6-4 victory at the first time of asking.
Serena was playing in her first tournament since recovering from the back problem that contributed to her fourth-round defeat by Ana Ivanovic at the Australian Open.
But she had looked relatively sharp in easing past Jelena Jankovic in the quarter-finals and the main doubt about a first all-Williams final since 2009 centred on whether Venus would beat Caroline Wozniacki.
The 33-year-old has found form of old in Dubai and won her 14th straight match at the tournament 6-2 6-3.
Venus was the champion in 2009 and 2010 and is through to her biggest final since being diagnosed with the auto-immune disease Sjogren's Syndrome in 2011.
The American said on wtatennis.com: "I feel like I've just gotten so much better since my first round, feeling comfortable on the court, feeling comfortable when I'm behind, feeling comfortable when I'm in the lead.
"It reminds me of when I first came on tour, just having to learn a lot, and I feel like I'm learning those lessons all over again.
"I had a lot of tough losses, and I realised I had to go through it. I felt like every time I got better.
"For me and for every player, you just have to keep stepping up to the line and believing in yourself."
