The French Open champion has not played since June due to a knee injury and opted out of the ongoing Australian Open when a virus disrupted his comeback plans.
Having already signed up to play in Brazil and Mexico during the South American claycourt swing, Nadal has now agreed to compete in the VTR Open in Vina del Mar.
The tournament is being held from February 4-10.
Nadal said: "I am delighted to confirm my participation in the tournament in Vina del Mar in Chile.
"I would like to thank the organisers of the tournament for this invitation, and say that I have many fans in the Andean country. Finally I can go in person to be with them and play my first tournament after the injury that has taken me away (for) so long."
Argentina's Juan Monaco, the world number 12, is currently the second-highest ranked player entered.
