The British number one claimed a famous win over Li in last year's third round at Flushing Meadows before losing to Sam Stosur in the last 16.
She has plenty of ranking points to defend so will aim to make the most of her first Grand Slam seeding. An opener against the 53rd-ranked Dominguez Lino should help, although the Spaniard has won both of the pair's previous meetings, most recently saving match point against the Briton in Katowice earlier this season.
If 30th seed Robson, who has been troubled by a wrist injury in recent weeks, is victorious she will meet either Caroline Garcia or Shelby Rogers in round two.
Meanwhile, the other Briton guaranteed a main-draw spot, Heather Watson, faces a tough test against 21st seed Simona Halep in her opening match.
The Romanian has won three WTA titles this season and only last week reached the quarter-finals in Cincinnati.
Top seed Serena Williams will open her US Open title defence against former French Open champion Francesca Schiavone.
Tougher tests lie in store further down the line with Sloane Stephens, who beat Williams at this season's Australian Open, on course to meet her fellow American in the fourth round.
Angelique Kerber is Williams' projected quarter-final opponent with Agnieszka Radwanska seeded to meet her in the last four.
On the other side of the draw, Cincinnati champion Victoria Azarenka is seeded to meet Sara Errani in the semi-finals. Azarenka may first have to battle past former Wimbledon champion Petra Kvitova in the last eight, while Errani's projected quarter-final is against Caroline Wozniacki.
Azarenka starts with a first-round match against Dinah Pfizenmaier.
