Vesnina was too strong for defending champion Barthel, wrapping up a 6-3 6-4 victory in one hour and 23 minutes to win the Australian Open lead-up tournament.
It was the 26-year-old's first WTA singles final, and the world number 68 broke down in tears after sealing her maiden trophy.
Vesnina had previously been haunted in tournament finals - she has lost six grand slam doubles deciders - in both women's and mixed - but struck at the first time of asking in her first professional tour singles final.
Both players struggled with their second serves in Hobart, with Vesnina winning just eight of 24 points behind her second ball, and Barthel 11 of 28, but the Russian did enough to see herself go into the year's opening major on a high.
Vesnina will meet French youngster Caroline Garcia in the first round at Melbourne Park, while Barthel - the 32nd seed - will play Kazakhstan's Ksenia Pervak.
