As expected, the seedings stuck rigidly to the world ranking list when they were annouced on Tuesday.
Five-time winner Roger Federer is therefore the top seed with defending champion Novak Djokovic seeded to meet him in the final.
Olympic champion Murray is the third seed - one place higher than his ranking due to Rafael Nadal's injury withdrawal. He will be hoping to follow his gold medal in London with a first Grand Slam title in New York, thus ending a 76-year wait for a British male singles winner at the highest level.
Spain's David Ferrer completes the top four seeds, while John Isner is the leading American seed at number nine.
The women's seedings are expected to be announced on Wednesday when it is likely Victoria Azarenka will be the top seed.
The draw for both singles events takes place in New York on Thursday at 1700 BST.
