The pair have been placed in the same section of the draw for the Sheffield event, which gets under way later this month.
After the world rankings were updated following Sunday's China Open final, the seeds were placed into the draw.
Despite not having played for months, O'Sullivan is top seed as he is the defending champion - he recently announced he would return to action at the World Championship. Higgins is a four-time winner, the last title coming in 2011.
World number one Mark Selby is in the bottom half of the draw and seeded to face Mark Allen in the last eight. A semi-final against China Open champion Neil Robertson could follow.
Meanwhile, O'Sullivan is seeded to face Judd Trump in the last four.
The seeds will be drawn against the 16 qualifiers on April 15. Qualifying takes place from April 4-14 with former champions Steve Davis and Peter Ebdon among those looking to earn a place in the Crucible field.
The World Championship starts on April 20 with the final on May 6.
