The French raider, trained by Andre Fabre and ridden by Maxime Guyon, tracked a sedate early pace and was produced to challenge passing the two furlong marker.
Ryan Moore made his move aboard 7/4 favourite Integral at the same time and for a stride or two looked set to pass the new leader.
However Esoterique willingly stuck to her guns and fended off the persistent challenge to score by a head, the pair well clear.
Fabre said: “She is a filly with a lovely temperament. Maxime had a tough opponent with Ryan Moore aboard.
“Her next race will be the Prix d’Ispahan at Longchamp and we will head to Deauville probably after that.”
Winning jockey Maxime Guyon added: “It rode like a good race. I had a good run behind the leader and she battled on before putting the race to bed. She will improve for the run as well. There’s a lot to look forward to.”
Connections of Integral were satisfied with her reappearance run and now all eyes turn to Royal Ascot.
“Ryan Moore just felt that things didn't quite pan out for him but you can't take nothing away from the winner. Integral has run a big race and they are two good fillies,” said Chris Richardson, managing director of Cheveley Park Stud, owners of Integral.
“She is as game as a pebble and I did think she got head back in front but it was not to be. All being well she will now go to Royal Ascot for the Duke of Cambridge.”
Paul Cole, trainer of Gifted Girl who finished third, felt that his horse was not suited by the pace in the race.
“We are very pleased with her and I think she has run well but she was not suited by the pace as it was a bit to slow for her,” said Cole.
“I said this morning that she would come on for the run but she was pretty much at her racing weight today. We will stick to these Group Three and Group Two races as she got better as the year went on last year.
“She is entered up at York but that may come a bit too soon while she is also in the Duke of Cambridge at Royal Ascot.”
