The ever-popular Italian, who will be riding in his 25th consecutive Arc and has won the great race three times, has come in for the mount with O'Brien's son, Joseph, unable to do the weight of 8st 11lb.
O'Brien junior instead rides stablemate St Nicholas Abbey.
Reporting Camelot in "good form" since his second place in the Ladbrokes St Leger that denied him the Triple Crown, O'Brien said: "Frankie is a great rider, he has all the experience in the world, and we're delighted to have him."
Ground conditions in Paris are soft, and O'Brien added: "We're hoping for not too much rain - we're hoping for English good to soft.
"He's in good form since Doncaster and the mile and six (in the St Leger) might have extended him too far there. We've always thought he was very special."
O'Brien is equally pleased with St Nicholas Abbey, who is returning to a mile and a half having run the last twice over 10 furlongs, most recently chasing home Snow Fairy and Nathaniel in the Irish Champion Stakes
He said: "I'm very happy with him. His preparation has gone brilliantly and he loves a mile and a half."
Mikel Delzangles believes Kesampour has several things in his favour despite being an outsider.
The French trainer was delighted with the three-year-old's run in the Prix Niel after a 15-week break and expects him to have come on for the outing.
Kesampour was only beaten a length and three-quarters by the winner Saonois in the Prix Niel over the course and distance, having finished just a length behind the same horse in the Prix du Jockey Club on his previous start in June.
"He is in very good form and I thought he ran a very good race in the Prix Niel after quite a long break," said Delzangles.
"I think he needed the race and hopefully he can improve. I was happy to see the rain as I think he will be a happier horse on the softer ground.
"He does not have a very good draw (stall 15), but we have to live with that. Hopefully he can run a good race."
