The two teams will tip off on January 15 in an NBA regular-season game, one of two next season to be staged outside the United States with the Houston Rockets facing the Minnesota Timberwolves in Mexico City in November.
While it will be the Bucks' first game on English soil, the Knicks enjoyed a 102-87 win over the Detroit Pistons at the same venue in January 2013.
Benjamin Morel, NBA senior vice-president and managing director, said: "We are proud to announce the fifth NBA regular-season game in London featuring the Milwaukee Bucks and the New York Knicks.
"Our NBA games in London have had a strong impact not just in the UK but also across Europe, and have proven to be a fantastic platform for our partners to connect with fans throughout the region."
League commissioner Adam Silver added: "While London and Mexico City each have their own unique culture, they both have rich sports traditions and share a great passion for the NBA.
"These regular-season games will bring the excitement and incredible competition of our league to more fans in Europe and Latin America."
Prior to the Knicks' previous visit, London hosted a 2011 double-header between the New Jersey Nets and the Toronto Raptors.
The Nets won both games, the second 137-136 after three overtime periods, and the relocated franchise returned this year under their new moniker of the Brooklyn Nets to beat the Atlanta Hawks 127-110.
The NBA Global Games programme for 2015 also includes pre-season games in Germany, Turkey, Brazil and two games in China.
