Maclin was carted off the practice field after going down untouched and the team later confirmed he had torn the ACL in his right knee.
The Eagles have yet to confirm Maclin's treatment schedule but he will require surgery and is likely to miss the entire season.
"He just came off the line of scrimmage, and his knee buckled a little bit," Maclin's fellow Eagles receiver DeSean Jackson said on the team's official website. "It's an unfortunate situation."
Maclin, 25, is entering his fifth season in the NFL and in the final year of his rookie contract.
He was the Eagles' leading receiver last season with 69 receptions for 857 yards and a team-high seven touchdowns.
Maclin had been expected to be a key feature of new Philadelphia head coach Chip Kelly's offence.
