Donaldson carded seven birdies and two bogeys in a five-under-par 66 at Valhalla to set himself up to gain some valuable ranking points after missing the cut in two of his last three events.
The 38-year-old currently occupies the final automatic qualifying place, but started the week less than two points behind Graeme McDowell in eighth and less than a point ahead of Luke Donald in 10th.
At eight under par, Donaldson was five shots clear of Donald and nine ahead of McDowell with 18 holes to play.
"It's for me to play well and take control and sort it out this week," Donaldson said. "I'm just trying to go out there and play as well I can and shoot as low a score as I can. It's one shot at a time and that's all I can do really. Let everything take care of itself.
"I'm in a decent position. I played really well all day. A few more putts could have dropped and it would have been a really strong position."
