The 23-year-old could yet link up with Roy Hodgson's England squad as they finalise preparations for the midweek Wembley clash.
But Spurs boss Sherwood believes the pacy full-back will not be ready for action, despite returning to club training following hip trouble.
Spurs midfielder Christian Eriksen will not recover from a back injury in time to line up against England either, Sherwood confirmed.
"Kyle won't be fit to play for England, he's not fit to play here today so he won't be fit for England either," he said.
"I'm not sure but he probably will [link up with England] as they will be training at our complex in the early part of the week.
"We're just hoping to nurse him back to fitness for next week, so we'll see how that goes.
"Christian Eriksen won't be ready for Denmark either by the way, after his back problem."
Walker's absence makes it almost certain Glen Johnson will start at right-back for Hodgson's men against Denmark, winning his 50th cap.
Sherwood said Walker was not ready to take part in Tottenham's edgy 1-0 Barclays Premier League victory over Cardiff at White Hart Lane on Sunday.
The former Blackburn midfielder is hopeful Walker will be available for Tottenham's Premier League trip to Chelsea on Saturday.
