Spurs seemed on course to finally finish above bitter North London rivals Arsenal last season, only to see the Gunners put an impressive run together to deny Andre Villas-Boas men a spot in the Champions League for this campaign.
And despite the obvious disappointment of missing out on a top four spot, Defoe believes that their summer transfer business has proved that they mean business this time around.
"Yes, people may say that we missed out on Champions League football last season," he told The Daily Mirror.
"But you look at the way the club has responded and straightaway you are excited about the way the team is being structured to go again."
Valencia hitman Roberto Soldado could seal a move to White Hart Lane in the next few days, and along with the additions of Paulinho from Corinthians and Nacer Chadli from FC Twente, Defoe is pleased with the way the squad is shaping up.
"You're talking about three top players who are going to take Tottenham to the next level," he added.
"Out of those three players, only Chadli has actually joined the squad so far but I've already told him the team spirit is incredible and that he will love it here."
"I can see why Spurs fans would be excited about Soldado coming because I am excited myself about the prospect of playing with him."
