The wicket-keeper, looking to win back his England place ahead of the international summer, has been struggling with an Achilles injury.
He started the season as a batsman only and, after missing two county matches, it was indicated he could fill that role again against the Red Rose.
Sussex have not named him in their travelling squad, though, citing his Achilles as the reason.
Prior is running out of time to convince new England coach Peter Moores to pick him for the first Test against Sri Lanka, which starts on June 12.
He was dropped after three Tests of the disastrous Ashes tour in favour of Yorkshire's Jonny Bairstow, but did score a hundred against Middlesex in April.
Bairstow himself has been injured, struggling with a broken thumb, but he is set to return against Durham on Sunday and will hope to showcase what he can do.
