Left-arm spinner Jadeja and seamer Mohit Sharma (3-22) did most of the damage as Kolkata finished short on 114-9 chasing a target of 149 in a match reduced to 17 overs by rain.
Robin Uthappa (47) and Yusuf Pathan (41) were the only Knight Riders to make double figures.
Opening batsman Brendon McCullum had earlier hit 50 off 40 balls to lead the Super Kings to a total of 148-3 after a delayed start in Ranchi.
The fixture was the first match of this year's tournament to be held on Indian soil. The opening fortnight took place in the Middle East due to elections.
The Super Kings sit second behind unbeaten Kings XI Punjab in the table on net run-rate, while the fourth-placed Knight Riders have lost four of their six matches and have played a game more than the other three teams on four points.
