Robin Uthappa (67) and Shakib Al Hasan (46no) powered the Knight Riders to a target of 155 with two overs to spare.
Uthappa put on 64 in 42 deliveries with opening partner Gautam Gambhir (21) but fell in the 12th over with 57 still required.
That was no problem for Shakib, who hammered six fours and two sixes in his 21-ball effort.
The Super Kings had finished on 154-4 after being sent in to bat, Suresh Raina's 52-ball 65 the top score.
The result gives fourth-placed Kolkata a four-point gap over their closest pursuers - Bangalore Royal Challengers and Sunrisers Hyderabad - for the final play-off spot.
Chennai have now lost two in a row but, with 16 points, sit second in the table behind Kings XI Punjab and are guaranteed to make the play-offs.
