Play was scheduled to begin at 2.30pm local time (1230 GMT) but heavy rain beforehand initially delayed that schedule and, despite a couple of brief interludes raising hopes of play, the rain returned each time and play was eventually called off just after 3.45pm local time.
The two teams will move on to Durban on Wednesday and then Centurion on Friday for the remaining two scheduled games.
Australia won the preceding three-Test series 2-1.
