Hampshire effectively ended Nottinghamshire's LV County Championship aspirations as Michael Lumb's career-best 219 put the visitors in position to chase an innings victory.
Lumb hit 37 fours from 325 balls and shared a fifth-wicket partnership of 208 from 237 balls with Sean Ervine, who himself made 104 for his third century of the season.
Hampshire eventually declared on 654 for eight after flogging the Notts bowlers to all parts of Trent Bridge, with Samit Patel recording figures of four for 199 from 48 overs.
Notts, needing victory to stay in touch with leaders Durham, collapsed to 48 for four at the close, with leg-spinner Imran Tahir claimin two wickets in two balls.
A brief rain shower delayed the start by 45 minutes, but Hampshire made their intent clear as soon as they resumed on 345 for three, as nightwatchman Tom Burrows square drove Charlie Shreck for four.
Burrows eventually clipped Shreck to Bilal Shafayat at midwicket for 20 but Ervine was quickly into his stride as he and fellow left-hander Lumb gathered runs freely, adding 71 in less than an hour before lunch.
After the interval the Hampshire batsmen were able to focus their attention on the record books, with a Lumb drive through the covers off Patel bringing up the 500 and passing his previous best of 172, set against Loughborough UCCE in May.
Notts captain Chris Read looked completely out of options with Shreck and Darren Pattinson providing no threat, and at one point medium-pacer Andre Adams opted to bowl off-spin in the afternoon session.
Lumb was finally dismissed by Patel with a ball that kept fractionally low as the batsman attempted to cut, and Ervine was caught at long-on off the occasional bowling of Shafayat two balls after reaching his hundred.
Dominic Cork and Imran Tahir were both lbw for nought as Patel picked up two cheap wickets before Hampshire skipper Dimitri Mascarenhas ended Notts' misery with a declaration after 161 overs in what was their fourth-highest total in history.
Things got even better for the visitors once Matthew Wood fended a short ball from David Griffiths at gully in the ninth over.
Tahir, who had picked up match-figures of 8-113 in last year's corresponding fixture, then had Mark Wagh and Pattinson lbw 11 overs later, and only missed out on a hat-trick when Lumb dropped Patel at leg-slip first ball.
Cork picked up the wicket of Scott Newman with a thin edge behind, and although Patel and Shafayat survived to the close, Notts are still 606 runs behind and need to reach 457 just to avoid the follow-on.
