The vacant British middleweight title will be contested by Billy Joe Saunders and John Ryder at the Copper Box Arena which hosted handball during the 2012 Games.
The first bout will be staged on September 21, with Warren revealing the venue should stage fights every two months in the 7,500-seat venue in Stratford.
"You have got Wembley which is 12,500 (capacity) and you have got nothing else down from that. This is half that capacity and a fabulous arena," Warren told BBC Sport.
London Legacy Development Corporation's Dennis Hone added: "Boxing belongs in east London and Queen Elizabeth Olympic Park will provide a powerful backdrop for the sport's heavy-hitters."
British Basketball League side London Lions are to play their home matches at the Copper Box from August and badminton's London Grand Prix is to be staged there in October.
