The American title-holder has apparently suffered a bicep injury in his training camp to scupper the scheduled February 23 fight in Detroit, Michigan.
The bout, originally set for January 19, has already been delayed once when unbeaten Sheffield fighter Brook injured his ankle in December.
Brook, the mandatory challenger, is currently away training in the Canary Islands in preparation for the bout.
American promoters Golden Boy have yet to announce the postponement officially but Brook's handlers, Matchroom, have confirmed the reports.
It is hoped Alexander will recover quickly and that the bout will only be delayed by four to six weeks. Matchroom boss Eddie Hearn is currently working on new dates with his American counterparts.
Brook said on Twitter: "Can't put into words how gutted I am."
